I want to put a TRS jack in my strat, with the ring connected to a momentary normally-open switch that closes to the sleeve (ground) when the switch is pressed. Then on my pedalboard I want to use this signal to control an effect (probably with a relay?)... Will this affect my tone in any way?

The switching may couple into the guitar signal as a pronounced "click", especially if both the tip and ring conductors are within the shield.
The coupling will depend on the interconductor capacitance, the loads, and the level of DC current switched.
